The ‘Spider-Man: Far From Home’ video release offers a tease of things to come. It sounds as though Sony is reviving the idea of putting the Sinister Six on the big screen. In one of the interviews on the disc’s bonus features, producer Amy Pascal teases:

“All of these villains that we now have in our universe happen to be characters that are in the Sinister Six. There may be something that happens with that.”
The Sinister Six has gone through multiple lineups, so almost all of Spider-Man’s major foes have been members at one time or another. The original roster, in ‘The Amazing Spider-Man Annual’ #1 (1964), consisted of Doctor Octopus (who assembled the team and acted as leader), Electro, Kraven the Hunter, Mysterio, Sandman, and the Vulture. So far, only the Vulture and Mysterio have appeared in the current cycle of movies, but Mysterio is dead, meaning that Vulture is the only one that could show up as a member.
Though he wasn’t a founder, The Scorpion has been part of the team, and Michael Mando appeared in ‘Spider-Man: Homecoming’ as the hoodlum who would eventually become The Scorpion, Mac Gargan, which sets him up to possibly return in the next ‘Spidey’ flick, alone or as part of this group.
Around 2013, prior to the release of ‘The Amazing Spider-Man 2’, Sony announced plans to craft an entire Spider-Man-based shared universe, with plans in place for ‘Venom’, a female-centered movie, as well as a heist picture featuring the Sinister Six. Drew Goddard (‘Daredevil’) was writing the screenplay, and was later announced to direct. The introduction of Electro (Jamie Foxx) in ‘Amazing 2’ was meant to set up ‘The Sinister Six’ and that picture even closed with the logos of various villains flashing across the screen during the closing credits.

But after ‘Amazing 2’ fizzled at the box office, Sony halted its existing plans. That’s when the studio struck its deal to share Spider-Man with Disney. The new Marvel-based Spider-Man, played by Tom Holland, with Jon Watts directing his solo films, was an instant hit with fans. Combined, the two new pictures, ‘Spider-Man: Homecoming’ and ‘Spider-Man: Far From Home’, have earned well over $2 billion, and ‘Far From Home’ is the highest-grossing ‘Spider-Man’ movie period. That’s what gave Sony the cajones to try and ditch Marvel Studios and its president Kevin Feige who had produced the pair. They figured the franchise was fixed after the dud ‘Amazing’ movies and they didn’t need Marvel’s help anymore.
The success of the two ‘Home’ movies also emboldened Sony to dust off their spin-off plans. Their first, ‘Venom’, was a hit. That further enforced Sony’s believe that they could do this without Marvel. Luckily that has since been reversed and Feige will return to direct the final (?) ‘Home’ flick.
Now, Pascal’s words imply that ‘Sinister Six’ is back in the works. It may be as a separate film, or maybe the plan is to bring a team of foes together in the third ‘Home’. We’ll have to wait and see how that goes.
For the record, there has been chatter that Sony is working on a solo ‘Kraven the Hunter’ movie, so that character is likely to be woven into ‘Sinister Six’. Venom is also a likely candidate.
